Realistically, how effective is CRISPR gene activation in mammalian cell line for the long term culture?
Hi all, I want to conduct a pilot for some spice in my project. I would like to upregulate certain intracellular protein/gene with CRISPR but considering if its even worth the time.
I hear wonders about CRISPR but mostly related to short term culture ... knockdown this or activate that for less than 3-4 passages. The transfection protocols seems pretty straightforward. Now, my particular application needs large culture numbers for several passages and I want to hear from you:
1. How effective CRISPR really is for protein/gene activation in mammalian cell lines for long term culture (say 20-30 passages)? 2. Does the selective antibiotics mess up too much the cells?
